Oligonucleotide Synthesis: The Basics

Summary: Learn the ins-and-outs of oligonucleotide synthesis.

Oligonuclotide synthesis is process used to create primers with methods like the replication of PCR. A relatively easy but useful process, Oligo synthesis has been a key part in the advancement of laboratory procedures. Chemical structures or nucleic acid fragments are synthesized in various sizes with an extreme focus on high purity. The purity levels are checked by a combination of liquid chromatography and spectrometry. Cleaning up the incomplete molecules and other components of the process are done through chromatography and are an important step. The products developed through synthesis are then analyzed using mass spectrometry and are quantified by UV spectrophotometry.

Customizing oligos have been a process that develops roughly 15 to 20 bases to limit how many sequences can be included within the primer sequence.

In molecular biology, DNA and RNA synthesis play huge roles. Oligonucleotides are one of the primary factors which operate roles such as sequencing, cloning and probes. For disease pathways, antisense therapy utilizes modified oligos to alter the key genes involved.

The synthesis process must be as accurate as possible to avoid errors. Molecular population, especially over certain values, should be carefully observed and users handling the process need to work with efficiency to deliver a successful project.

With continuous and exponential growth within molecular biology, new types of methods are being produced that could potentially enhance the field of scope that scientists and biologists are working in. The growth factor within this industry continues to develop prototypes of mechanisms that will give a whole new perspective in regards to synthesis.
